Dr. Jansen's major basic research area is in protein nutrition, with much but not all of it involving protein quality/quantity/energy relationships during lactation. His research model, investigated with his research associates and graduate students, involved feeding wheat protein with and without supplementary amino acids at two protein and two energy levels to rats during pregnancy and lactation with comparison to methionine supplemented casein. In this work, protein synthesis, serum and tissue amino acids, mammary gland development and brain development in the pups were studied. This led to 18 research papers published mostly in the Journal of Nutrition as well as a number of book chapters and reviews.
These papers emphasize the importance of protein quality, i.e. the dietary supply of indispensable amino acids, in controlling protein synthesis during lactation. Absolute protein synthesis rate in the mammary gland was nearly twice as high in casein than in wheat fed dams fed protein at 11% even when pair fed at 85% of the ad libitum consumption of the wheat fed dams. Changes in mammary protein synthesis and pup weight were highly correlated with the protein nutritional value of the diet. Protein synthesis in the liver was less affected than protein synthesis in the mammary bland by poor protein quality. Under conditions of the experiments, a direct relationship between dietary lysine, free lysine in the mammary gland and milk secretion was observed.
Free amino acid concentration in the brain were highly correlated with serum levels for all 16 amino acids measured. Pup growth was five times higher in dams fed wheat supplemented with lysine and threonine at 21% compared with that observed in dams fed unsupplemented wheat protein at 11%. Under these conditions the concentration of lysine and threonine in the brain were twice as high in the former as compared to the latter conditions, with the differences in other amino acids less than 20%.
In the above experiments wheat protein was supplied by white bread. When 13% wheat gluten was fed during gestation and lactation, an almost total failure of lactation resulted. This was attributed to a reduction in glucose utilization per secretory cell and reduction in mammary gland size and content of DNA, RNA and protein
The interaction of dietary protein with the environmental stresses of heat and crowding were studied in a 1991 paper published in Physiology and Behavior. Many interesting relationships were elucidated in this study. The bottom line is that lactating rats have considerable ability to raise successfully their litters under adverse environmental conditions.
In addition to the above referenced papers, Dr. Jansen has published many papers over his career demonstrating the value of amino acid fortification and the importance of protein quality, including landmark reviews published as a supplement to the Journal of Nutrition (1962), and as chapters published by Academic Press in 1974, 1977 and 1982.
Another basic research area concerned obesity and lipogenesis. A model was developed at the Merck Institute to study lipogenesis in the mouse in vivo leading to a series of 10 papers published mostly in the Biochemical Journal and Archives of Biochemistry and Biophysics in the period 1966-1970. These papers contributed significantly to the knowledge base in the late 1960's on the dietary control of lipogenesis. Other papers explored obesity in rodent models and sites for the synthesis of cholesterol. A paper published in the American Journal of Physiology in 1969 was among the first to demonstrate that hypothalamic obesity in the rat could be produced by knife cuts around the ventromedial nuclei. A paper published in Archives of Biochemistry and Biophysics in 1970 was among the first, if not the first, to demonstrate and quantitate the importance of skin as a site for cholesterol synthesis in vivo.
The most significant research carried out in obesity at Merck was using obob mice and reported in the Biochemical Journal in 1967. When obob mice were fed a purified diet containing 1% corn oil and no cholesterol for 12 weeks, the liver contained 1.5% cholesterol and 20% fat. This was not true for the non-obese littermates. Cholesterol synthesis was reduced. However when [U-14C]-glucose was fed in the diet over a 24 hour period, there was a very large accumulation of labeled cholesterol in the liver suggesting that the accumulation resulted from a difficulty in removal of newly synthesized cholesterol from the liver. In follow-up work carried out at that time at Merck, but published in 1996 in the Journal of Nutritional Biochemistry because of the explosion of interest in the obob mouse, the accumulation of cholesterol in the liver in the obob mouse fed a 1% corn oil diet was confirmed. It was further demonstrated that this accumulation did not occur in gold thioglucose obese mice. Most importantly, it was demonstrated that feeding the bile acid analog D22-5B-taurocholenic acid reduced liver cholesterol to normal suggesting that the accumulation is related to interactions among cholesterol, bile acids and phospholipids in bile secretion. Following up these observations is currently the subject of a research proposal to the Competitive Research Grants Program of the USDA in collaboration with a colleague Dr. Kenneth Allen
In the applied nutrition area Dr. Jansen with a colleague, Dr. Jud Harper, in the Chemical Engineering Department carried out extensive research programs involving the national school lunch program and the processing of low-cost weaning foods in developing countries. This led to the publication of approximately 24 research articles and reviews. The low cost weaning food project led to USDA/AID implementation consultancies in Sri Lanka, Tanzania, Ecuador, Egypt, India and the Sudan.
In the nutrition education area the major innovation was the development and evaluation of nutrient approach to planning diets meeting prudent diet goals. This effort led to the publication of 8 research articles and a Prudent Diet Planning book published in 1990 by Academic Press.
Dr. Jansen is a member of the American Society for Nutritional Sciences, the American Society for Biochemistry and Molecular Biology, Institute of Food Technology, AAAS, Sigma Xi and Phi Kappa Phi. He has been a member of the Editorial Board of the Journal of Nutrition and several other journals, Program Manager of the USDA's Competitive Grants Program in Human Nutrition, NAS Committee on Evaluating Pre-doctoral Fellowship in Biomedical Sciences, (including chair), the USDA's Board of Scientific Counselors in Human Nutrition, Executive Committee of the IFT, and remains a member of the Committee on Military Nutrition of the IOM/FNB and on the Scientific Advisory Board of the American Committee on Science and Health.
Dr Jansen received the USDA Superior Service Award for Superior
Service in Research for his work in protein nutrition during lactation,
the Babcock-Hart Award of the IFT, and a Certificate of Merit from the
OICD/USDA for work in weaning food development and diet planning, awards
from the Colorado Dietetic Association, Western Dairy Council, and the
Colorado Affiliate of the American Heart Association for his work in nutrition
education, and the Distinguished Service Award from Colorado State University
for leadership at the Department Head level. He is a Fellow of the
American Society for Nutritional Sciences (1998) and the Institute of Food
Technologists (1990)
